P-1: Morphine Has Detrimental Effects on Sperm Parameters, Chromatin Quality and DNA Integrity in Mice
Authors
Abstract:
Background Morphine as a natural alkaloid (opiate) is considered as one of the problems associated with poor semen production and sperm quality. For the first time, this experimental study was carried out to evaluate the impact of intra-peritoneal injection of morphine on sperm parameters, chromatin quality and DNA integrity of spermatozoa aspirated from cauda epididymis of mice. MaterialsAndMethods Totally 24 adult male balb/c mice (8 weeks old. 30 g) were equally divided into 3 groups (n=8). Mice of group 1 served as control fed on basal diet, group 2 received basal diet and normal saline and group 3 received basal diet and morphine (15 mg/kg/daily, intraperitoneal) for 35 days. Finally, the right tail of epididymis of each animal was removed and placed in Ham’s F10 for 30 min. Released spermatozoa were used to analyze count, motility, viability (eosin-nigrosin staining), morphology (Papanicolaou stain), chromatin quality with Toluidine blue (TB) and apoptosis via TUNEL assay. Results In morphine-treated mice a significant decrease was found in sperm viability, normal morphology, count and motility compared to other groups (P

P-61: Effects of Acrylamid on Sperm Parameters and Chromatin Quality in Mice
Background: Acrylamid (AA) is found in carbohydraterich foods that are prepared at high temperatures, such as French fries and potato chips. The toxic effects of AA in male animals include degeneration of epithelial cells of seminiferous tubules, decreased number of sperm and elevation in abnormal spermatozoa. The aim of this study was to evaluate the detrimental effects of AA on sperm paramete...
full textAdministration of high dose of methamphetamine has detrimental effects on sperm parameters and DNA integrity in mice
BACKGROUND Methamphetamine (MA) was shown to have harmful effects on male reproductive system. OBJECTIVE To investigate probable effects of daily administration of MA on sperm parameters and chromatin/DNA integrity in mouse. MATERIAL AND METHODS Thirty-five NMRI male mice were divided into five groups including low, medium, and high dosage groups which were injected intraperitoneally with 4...
full textP-12: The Improvement of Sperm Parameters and Chromatin Quality by Vitamin E Supplement in Mice
Background: Vitamin E is believed to be the primary component of the antioxidant system in spermatozoa. There is a relationship between activity of these antioxidant and function of sperm. Vitamins E and C which are belong to non-enzymatic antioxidant are used as a supplemented drug to improve sperm quality and male fertility. The present study was carried out to investigate the positive effect...
full textearly and late effects of ibuprofen on mouse sperm parameters, chromatin condensation, and dna integrity in mice
background: there are few studies indicating the detrimental effects of ibuprofen on sperm fertility potential and dna integrity. objective: to determine the effects of ibuprofen on sperm parameters, chromatin condensation and dna integrity of mice. materials and methods: in this experimental study, 36 adult male mice with average weight 37 gr were divided into three groups, including control (...
full textP-235: Ceftriaxone Effects on Sperm Quality and DNA Damage in Adult Mice
Background: Ceftriaxone is a third-generation Cephaloporin antibiotic. It has broad spectrum activity against gram-positive and gram-negative bacteria. There are some reports about its adverse effects on sperm quality and quantity but there is a need for more investigations so we did it. Materials and Methods: 40 adult male mice were divided into 3 groups as followings: a control group, high do...
full textAdvancing age has differential effects on DNA damage, chromatin integrity, gene mutations, and aneuploidies in sperm.
This study compares the relative effects of advancing male age on multiple genomic defects in human sperm [DNA fragmentation index (DFI), chromatin integrity, gene mutations, and numerical chromosomal abnormalities], characterizes the relationships among these defects and with semen quality, and estimates the incidence of susceptible individuals for a well characterized nonclinical nonsmoking g...
